چکیده

Ginkgolides A, B, and C are diterpene trilactones and active constituents of the 50:1 Ginkgo biloba leaf extract widely used in the symptomatic treatment of mild to moderate dementia. Using the two-electrode voltage clamp methodology, these ginkgolides were found to be moderately potent antagonists at recombinant human a1h2g2L GABAA receptors expressed in Xenopus oocytes. Ginkgolides A, B, and C inhibited the direct action of g-aminobutyric acid (GABA) with Ki values of 14.5F 1.0, 12.7F 1.7, and 16.3F 2.4 AM respectively. Antagonism by these ginkgolides at a1h2g2L GABAA receptors appears to be noncompetitive as indicated by the nonparallel right shift and reduced maximal GABA response in their GABA concentration–effect curves. Terpene trilactones from Ginkgo biloba are antagonists of cortical glycine and GABA(A) receptors.

Glycine and gamma-aminobutyric acid, type A (GABA(A)) receptors are members of the ligand-gated ion channel superfamily that mediate inhibitory synaptic transmission in the adult central nervous system. During development, the activation of these receptors leads to membrane depolarization. Ligands for the two receptors have important implications both in disease therapy and as pharmacological t...

Mixed antagonistic effects of the ginkgolides at recombinant human ρ1 GABAC receptors

The diterpene lactones of Ginkgo biloba, ginkgolides A, B and C are antagonists at a range of Cys-loop receptors. This study examined the effects of the ginkgolides at recombinant human ρ(1) GABA(C) receptors expressed in Xenopus oocytes using two-electrode voltage clamp. The ginkgolides were moderately potent antagonists with IC(50)s in the μM range. At 10 μM, 30 μM and 100 μM, the ginkgolides...

Chemistry and biology of terpene trilactones from Ginkgo biloba.

Ginkgo biloba, the ginkgo tree, is the oldest living tree, with a long history of use in traditional Chinese medicine. In recent years, the leaf extracts have been widely sold as phytomedicine in Europe and as a dietary supplement worldwide. Effects of Ginkgo biloba extracts have been postulated to include improvement of memory, increased blood circulation, as well as beneficial effects to suff...

Terpene trilactones from Gingko biloba: from ancient times to the 21st century.

Ginkgolides were always close to my heart. I continue to be strongly attracted to Ginkgo biloba, the ginkgolides and bilobalide. Starting in 1963, I became fascinated by these molecules while working on their isolation and structure elucidation in Sendai. Presumably, due to the ginkgolide studies, I received an invitation to join the faculty at Columbia University. After almost three decades of...

Ginkgolide derivatives for photolabeling studies: preparation and pharmacological evaluation.

The terpene trilactones (TTLs), ginkgolides and bilobalide, are structurally unique constituents of Ginkgo biloba extracts, which exhibit various neuromodulatory properties. Although the TTLs are believed to be responsible for some of these effects, the specific interactions with targets in the central nervous system remain to be elucidated on a molecular level. Ginkgolides are known antagonist...
